Clear Test Amphetamine Surestep is a test for the quantitative determination of amphetamines in human urine. The test in its initial stage is immunochromatographically based on the in vitro test. The presence of amphetamine in human urine at a cut-off level of 1000 ng/ml can be detected in 3 minutes. The amphetamine test is a qualitative analysis. It identifies d-amphetamines in human urine at a concentration of 1000 ng/ml or higher. The concentration of d-amphetamine cannot be determined by this assay. The test is designed to distinguish a negative result from a presumed positive result. All positive results should be confirmed using an alternative method, preferably GC/MS. The amphetamine test is based on the principle of the specific immunochemical reaction between antibodies and antigens to analyze the specific components in human urine specimens. The quantitative assay is based on the competition for antibody binding between a drug compound and a free drug that may be present in the urine sample being tested. When a drug is present in the urine sample, it competes with the drug compound for the limited amount of the antibody-stain compound. When the amount of drug is equal to or more than 1000 nl/mg, it will prevent free drug from binding to antibodies. Therefore, a positive urine sample will not show a colored band in the test area and give a positive result, while the presence of a colored band indicates a negative result. There is a control line on the test that serves as a procedural control. This colored band should always appear in the control area if the test is stored under good conditions and performed correctly. Amphetamines are a group of strong sympathomimetics with therapeutic applications. The most common of the amphetamines are d-amphetamine and d,1-amphetamine. They are mainly stimulants of the central nervous system, causing neurotransmitters and the release of epinephrine, norepinephrine and dopamine in the brain and body - leading to euphoria, excitement with a feeling of energy, self-confidence and mobility. Chronic abuse of amphetamines leads to tolerance and habituation to the opiate effect. The cardiovascular system responds to amphetamine with an increase in blood pressure and arrhythmia. It also causes anxiety, feelings of fear, paranoia, hallucinations and psychopathic behavior. Amphetamine is metabolized by many pathways. To a greater extent, acidic urine helps excretion, while alkaline urine prevents it. In 24 hours, approximately 79% of amphetamine is excreted in acidic urine and about 45% in alkaline urine. How to use the Clear Amphetamine Test Strip.
Sample collection and preparation:
A sample of approximately 150ml is required for each test. Fresh urine samples do not need special handling or processing. The sample should be taken in a clean, dry, plastic or glass container. If the quantitative test is not performed immediately, the urine sample can be cooled to 2-8 C and stored for up to 7 days or frozen. Samples should reach room temperature before testing. Urine samples that show a large sediment or turbidity should be centrifuged or allowed to settle so that a clear liquid is obtained for the test.
Quality control:
The control strip is an internal reagent and procedure control. It will appear if the test was performed correctly and the reagents were reactivated.
Control standards can be used to validate reagent performance and establish test reliability. Controls not provided with this test may be purchased.
Procedure for the test strip:
1. Allow all materials and samples to reach room temperature.
2. Remove the test strip from the sealed foil.
3. Dip the strip into the urine sample so that the arrows point to it. The sample level should not be higher than the maximum indicated by an arrow.
4. Hold the strip in the urine until a colored pink line appears in the test area (approximately 20 seconds).
5. Remove the strip and place it face up on a clean, non-absorbent surface or leave it in the urine if the sample level is not higher than the maximum indicated by an arrow.
6. Read the result between 3-8 minutes.
Do not interpret a result after 10 minutes!
Interpreting the results:
Negative: Two colored bars appear. The appearance of two colored bands, one in the test zone line and one in the control line zone, indicates negative results. A negative result does not indicate the absence of amphetamines in the specimens. It only indicates that the level of amphetamines in the samples is below the cut-off level.
Positive: A colored band appears in the control line area. No colored band is detected in the test line area. This is an indication that the level of amphetamines in the urine specimen is above the permissible limit.
Invalid: If there is no colored band in the control line area – the test result is invalid. Retest the sample with a new test device.
Note: A very light colored band in the test line area indicates that the amount of amphetamines in the sample is close to the legal limit level. These specimens and other positive samples should be confirmed using an alternative method such as GC/MS.

Do not open the sealed foil until you are ready for the test.
Use a new container for each urine sample to avoid contaminating one sample with another.
The test kit should be stored at 2 to 30 C and is effective until the expiration date.
